These are from the DNA that cae out of my mini-prep. I decided to increase my chances of success by taking two samples from the plates and growing them over night and mini-preping them. All four samples, two form otsA and two from otsB, were digested with AlwnI and BglI. the two on the left show that the plasmid was cut at around 1600bp, 1200bp, and 600bp which was what was expected if the plasmid contained otsA. The two bands on the right are 1200bp, 1000bp and 600bp which was expected if the plasmid contained otsB. Cloning was a success!!
