5:45 Status Reports: (5 minutes each including any discussion)

  • Alisa: Ligation experiments progress.
    • Problem: 1.2KB genes between biobrick forward & reverse primers not being visualised in colony PCR with 1.5min extn time. (300bp fragments do amplify)
      • - craig same ligation worked fine [no BSA, no gel purification, no Alkaline phosphatase used]
      • - craig very high 10mg/ml DNA yield from miniprep using alkaline lysis technique.
      • - Alisa: doesn't believe it is worth investigating any further except as experiments are done.
  • Pat & Alisa: Red light receptor progress/design/plan- needs?
    • phase 1 osmolarity test of YFP under enz
    • phase 2 Red light driving YFP
    • Phil would like to see lacZ coliphotography parts available for tests due to higher sensitivity of enzyme output.
  • Jan: Blue light receptor design/plan - needs?
    • Phil sugests using prism for optimal colour discovery before ordering filters, Projector as source.
    • Review to be cionducted with Jan, Paul & Alisa
    • No need to order any apperatus yet.
  • Phil: Gas vesicles progress/design/plan - needs?
    • Plan up to basic biobrick complete,
    • First mutagenesis in progress using complementry primers hand designed, and one overlapping and one computer generated using stratagene program.
    • Plan for support parts/test harness required.
    • Lac promotor and antibiotic death plasmids required.
  • Kasia: Documentation progress vs last report/ new problems/ needs.-No report

    • Priority should be on plans and experimental report analysis and conclusions-----Alisa .
    • If only Pat & Alisa doing lab work not much documentation needed.
    • Equipement:
      • MissingLas3000 method (in lab notebook)
      • Missing DNA measurement protocol (eppendorf)
    • Reagents pages incomplete:
      • Some location pictures not finished or uploaded.
      • Some newer reagents not entered.
