Engineering Escherichia coli to see light

To create the chimaera, we aligned members of the phytochrome family with EnvZ and identified potential functional crossover points between the Synechocystis phytochrome Cph1 and EnvZ. The length and composition of the peptide that links a photoreceptor to its response-regulator can affect signal transduction, and we therefore constructed a series of chimaeras with variable linker lengths. The variants were transformed into a <img alt="Delta" align="bottom" border="0" src="http://www.nature.com/__chars/_delta/black/med/base/glyph.gif" />EnvZ E. coli strain containing a chromosomal fusion between the OmpR-dependent ompC promoter and the lacZ reporter, which enzymatically produces a black compound.

  1. The active chimera contains the first 517 a.a. of Cph1 and he last 229 a.a. of EnvZ

  1. Most receptors in E. coli located in the cytoplasmic membrane and share similar transmembrane topology.
  2. The N-terminal extracytoplasmic domains are flanked by two transmembrane domains.
  3. The second transmembrane domain is connected to the cytoplasmic region designated as a "linker", which is followed by the cytoplasmic signaling domain.
  4.
