- 1. Mix :
- -1 μg of plasmid
- -3 μl of Buffer 10X
- -0.3 μl BSA 100X
- -3 units of restriction enzyme
- -water up to 30 μl
- 2. Incubate at 37 °C for 1 hr.
- 3. Incubate at the inactivation temperature of the enzyme for 20 minutes.
