In flow cytometry the values obtained from filters donot exactly give protien values. If there is some overlap between protien emission spectrum and filter range then protien gives signal in that filter. We came up with a mathematical method to seperate CFP and YFP from autofluorescence and noise. The details about this mathematical tool for correction can be found here.[[Media:Analysis.pdf|here]].

The following is the record of all the experiments done by us, each followed by graphs obtained by analysis of the corresponding microscopy and flow cytometry data.

In flow cytometry the values obtained from filters donot exactly give protien values. If there is some overlap between protien emission spectrum and filter range then protien gives signal in that filter. We came up with a mathematical method to seperate CFP and YFP from autofluorescence and noise. The details about this mathematical tool for correction can be found here.here.
