Experiments

Microscopy:

  • Open loop trial - 50 ng/ml aTc
  • Full range induction of pL CFP for equivalence. NOT final because OD was >0.1 (~0.4-1.0)

  • Open loop trial - 50 ng/ml aTc
  1. For both cases of AI (grown at 50 ng/mL and ODs 0.2/2), make Scatter plots of CFP vs LuxR.YFP for all 6 concentrations of IPTG.
  2. A combined plot of Mean and S.D of CFP and LuxR.Y having points from OD 0.2 in one colour and OD 2 in the other colour.

Inference

aTc being an antibiotic may have adverse effects on the cell at high concentrations such as 100 ng/ml. Growth rate is reduced and cells are killed. These factors may in turn affect the expression of the fluorescence gene which may lead to a drop in mean fluorescence intensity at high concentration levels.
