Ligation
Use Nanodrop to find vector and insert concentration in [ng/uL]. Find the length of vector and insert and calculate volume of reagents needed with [http://mywebspace.wisc.edu/klapoetke/web/iGEM/WIDGET_Ligation_Reaction.xls this excel spreadsheet].
Heat inactivate ligation afterwards: 15 minutes at 65C.
