- 'Saccharomyces cerevisiae strain CB011: W303 MATa, Ste5::KanR, bar1::NatR, far1D, mfa2::pFus1-GFP, his 3, trp1, leu2, ura3 was used to integrate each synthetic scaffold (pSte5-Ste5-EE-Adh Terminator or pSte5-Ste5-RR-Adh Terminator) into the HO locus.
- Subsequently, each pRS315-based construct was transformed (selection based on Leu2 marker). Note that Negative Effectors fused to EE zippers will be recruited only to Scaffolds fused to RR zippers, whereas Negative Effectors fused to RR zippers will be recruited only to Scaffolds fused to EE zippers.
