To Do List:
1a- start restriction on mini'd Benny in Boo samples (single digest should be sufficient)
1b- all we are looking for here is an increase in size the size of Benny (you can do a double digest Eco/Pst which will directly show us the band size (but will take longer...)
1c- run a gel of the restricted products to show our fate - we hope to see an increase in ligated samples compared to the Boo34 restricted control (we made 0.75% gel w/ EtBr last nite)
2- complete mini's on Enny in Boo overnites (we started at 9pm)
3- Overnites of the newly transformed genes Buddy and Betty! which are in 37degrees
4 - Autoclave test tubes from the dishwasher
5 - if needed, purify newly restricted boo34 (spe/pst) via gel (tape half of comb and add all restricted product)
6- Fridge is fixed
