- phase 1 osmolarity test of YFP under enz
- phase 2 Red light driving YFP
- Nothing planned for this week due to academic load.
- Jan: Blue light receptor design/plan - needs?
- Jan will talk to physics about prism, nothing else planned this week
- Phil: Gas vesicles progress/design/plan - needs?
- Plan up to basic biobrick complete,
- First mutagenesis complete: 4 changes sucessfull independently , 4 unsucessfull. Unsuccesssfull changes correlated with Tm of primers will repeat with longer melting periods. Test of primer design systems yeilded varying results, Best(30colonies: 2/3 good) was stratagene program followed by jeremy's(Wehi offset design)(13 colonies 0/3 good) followed by steves (complementary hand design to stratagenes rules)(3 colonies 1/3 good). On this basis Phil had entered statagene designed primers on geneworks order form, Alisa to submit for Friday delivery.
- Need to check expression of gas vesicles Chris to perform.
- Plan for support parts/test harness required.
- Lac promotor and antibiotic death plasmids required.
- Kasia: Documentation progress vs last report/ new problems/ needs.-No report

- Ligation protocol missing
- Lab notebook:
- Format uses too many lines to see connections between days.
- Latter part missing links
- Can table of contents be restricted to date level?
- Experimental reports:
- Lacks discussion/conclusion
- Ligation experiment missing
- Lattest few gells not marked up and entered.
