• Dilutions of pT luxI.cfp made for equivalence.

• pT.luxI.cfp inoculated into LB for Open Loop[A] (morning)

• pT.luxI.cfp and K12Z1 inoculated in LB for Open Loop[B] (evening)

• pLac.cfp Inoculated for equivalence.

• Inoculated into 50 mL Glu M9 with 20 ng/mL aTc (20*50=1 ug == 100 uL of 10 ug/mL)

            pT LuxI.C: 2 flasks - 1 ul/mL == 50 uL  

[A] - 0.1 ul/mL == 5 uL (Krishna)

            K12z1:     1 flask  - 2 ul/mL == 100 uL 

• Inoculated pL luxR yfp.pR cfp and K12Z1 in LB for Open Loop[A]

• pLac.Cfp dilutions done (14+2+4=20 tubes)

Microscopy

• OD was measured and pT luxI.C imaging done for equivalence.

• pL.luxR.yfp imaging done for equivalence.
