** Problem: 1.2KB genes between biobrick forward & reverse primers not being visualised in colony PCR with 1.5min extn time. (300bp fragments do amplify)
+
** Attempting ligation of RBS to LacZ reporter.
-
*** - craig same ligation worked fine [no BSA, no gel purification, no Alkaline phosphatase used]
+
-
*** - craig very high 10mg/ml DNA yield from miniprep using alkaline lysis technique.

-
*** - Alisa: doesn't believe it is worth investigating any further except as experiments are done.

** First mutagenesis complete: 4 changes sucessfull independently , 4 unsucessfull. Unsuccesssfull changes correlated with Tm of primers will repeat with longer melting periods. Test of primer design systems yeilded varying results, Best(30colonies: 2/3 good) was stratagene program followed by jeremy's(Wehi offset design)(13 colonies 0/3 good) followed by steves (complementary hand design to stratagenes rules)(3 colonies 1/3 good). On this basis Phil had entered statagene designed primers on geneworks order form, Alisa to submit for Friday delivery.

Documentation wiki status:

    • Priority should be on plans and experimental report analysis and conclusions-----Alisa .
    • If only Pat & Alisa doing lab work not much documentation needed.
    • Equipement:
      • MissingLas3000 method (in lab notebook)
      • Missing DNA measurement protocol (eppendorf)
    • Reagents pages incomplete:
      • Some location pictures not finished or uploaded.
