5:30 Action points from last meeting:

  • Alisa& Pat: Wiki "Parts_used status" up to date? -(outstanfding from 19/7/07)
  • Jan: Lecture Timetables spread sheet to be emailed to all->on google groups
  • Pat & Alisa: Plan for red light receptor on wiki -(outstanding from 3/8/07)
  • Alisa, Paul & Jan: Review blue light theory - done
  • Jan:Theory for blue light on wiki-done
  • Jan:ComP fusion to be ordered.- 31 August shipping
  • Jan:Production of sequence for blue light receptor? " -designed
  • Jan:Plan for blue light on wiki by 17th August- Done (needs more details- coundn't implement this)
  • Jan:<b/>Order red and blue filters.-dropped
  • <b>Paul: Status of #BBa-M30109 & #BBa-V1012 lost in transit?- Phil to chase (one unmarked tube arrived)
  • Craig: Alkaline lysis technique for miniprep to be documented
  • Cheng: Daisy to produce competent cells of DB3.1 and Enz- strains.- waiting on strains still

5:45 Status Reports: (5 minutes each including any discussion)

  • Alisa: Ligation experiments progress.
    • Attempting ligation of RBS to LacZ reporter.
  • Pat & Alisa: Red light receptor progress/design/plan- needs?
    • phase 1 osmolarity test of YFP under enz
    • phase 2 Red light driving YFP
    • Nothing planned for this week due to academic load.
  • Jan: Blue light receptor design/plan - needs?
    • Jan will talk to physics about prism, nothing else planned this week
  • Phil: Gas vesicles progress/design/plan - needs?
    • Plan up to basic biobrick complete,
    • First mutagenesis complete: 4 changes sucessfull independently , 4 unsucessfull. Unsuccesssfull changes correlated with Tm of primers will repeat with longer melting periods. Test of primer design systems yeilded varying results, Best(30colonies: 2/3 good) was stratagene program followed by jeremy's(Wehi offset design)(13 colonies 0/3 good) followed by steves (complementary hand design to stratagenes rules)(3 colonies 1/3 good). On this basis Phil had entered statagene designed primers on geneworks order form, Alisa to submit for Friday delivery.
    • Plan for support parts/test harness required.
    • Lac promotor and antibiotic death plasmids required.
  • Kasia: Documentation progress vs last report/ new problems/ needs.-No report

    • Priority should be on plans and experimental report analysis and conclusions-----Alisa .
    • If only Pat & Alisa doing lab work not much documentation needed.
    • Equipement:
      • MissingLas3000 method (in lab notebook)
      • Missing DNA measurement protocol (eppendorf)
    • Reagents pages incomplete:
      • Some location pictures not finished or uploaded.
      • Some newer reagents not entered.
    • Secondary reagents pages not made:
      • LB, Plates, TAE , NZY+ Broth
    • Protocol part:
      • No connections from Protocols to reagents.

      • Format uses too many lines to see connections between days.
