Digestion of PCR product or Miniprepped DNA (50 uL scale):

       42.1 uL PCR product or miniprepped DNA

       5.0 uL appropriate NEB (10x)

       0.5 uL BSA (100x)

       1.2 uL Enzyme 1

       1.2 uL Enzyme 2

       50 uL total

• Incubate at 37ºC for 2 hours

• Add 0.5 uL of each enzyme

• Incubate at 37 ºC for an additional 30 min.

• Run gel for gel extraction
