1. Mix :

- 1μg plasmid

- 3μl Buffer 10x [ ]

- 0.3μl BSA 100x [ ]

- 3 units of restriction enzyme

- water up to 30μl

2. Incubate at 37 °C for 1hr.

3. Incubate at the inactivation temperature of the enzyme for 20min.
