(final volume 30μl)

1. Mix :

- 1 μg of plasmid

- 3 μl of Buffer 10X

- 0.3 μl BSA 100X

- 3 units of restriction enzyme

- water up to 30 μl

2. Incubate at 37 °C for 1 hr.

3. Incubate at the inactivation temperature of the enzyme for 20 minutes.
