Experiments

For [C]

• The inductions were done in 50 mL Glu M9 in 250 mL flask with 5 ng/mL conc of aTc and 1,0.1 uL/mL of the inoculum. (5*50 ng=0.25 ug == 25 uL of 10 ug/mL aTc)

• pLLuxR.Y.pR.C and K12z1 were inoculated in LB.

• Filter required OD flask, store at 4 degrees.

• Seven dilutions for pL.LuxR.Y.pR.C were made with 50 old:50 new 2xM9 along with -ve control: K12Z1.

For [D]

• The inductions were done in 50 mL Glu M9 in 250 mL flask with [aTc]=1 ng/mL and inoculum=1,0.1 uL/mL (1*50 ng=0.05 ug == 5 uL of 10 ug/mL aTc)

• 0100: Make 7 dilutions for pL.LuxR.Y.pR.C with 50 old:50 new 2xM9
