Hello Tim,

<u>'''Quorum-sensing coupled with the Repressilator'''</u>

we need a lab notebook page, protocols page and team members page, etc.

Our project is a continuation of one of the projects we presented last year: a two-gene oscillator with an 'On' and 'Off' switch using LacI and LuxI. PS: Everyone, get a lab notebook! We're making our cells chemically competent tomorrow so if you want to see/help out then show up around 10:30. [[User:Horiavulpe|Horiavulpe]] 16:41, 14 May 2007 (EDT)

'''Team 1 - Fluorescence Complimentation'''

-
This project on fluorescence complementation involves confirming previous work using Jun and Fos heterodimerizing proteins, each fused to a half terminus of YFP. The two halves of the YFP protein bind, giving rise to fluorescence.  We will then carry out the experiment with GFP or YFP split at different points in order to find a cleavage point that produces reversible fluorescence.  These half proteins will then be attached to proteins that revesibly associate.<br>

Our project consists of a two-gene oscillator with coupling by an Autoinducer (AI). This system was presented last year. We will test the effects of adding AI and Tetracycline to the medium. We will also investigate the nature of the oscillations at different cell densities.<br>

Project Overview

Quorum-sensing coupled with the Repressilator

Our project is a continuation of one of the projects we presented last year: a two-gene oscillator with an 'On' and 'Off' switch using LacI and LuxI. This simple relaxation oscillator could be used as a modular component for the synchronization of other complex oscillating systems such as the repressilator.

Quorum-sensing coupling between cells is achieved with a diffusible autoinducer AI (AHL) made from LuxI which promotes the synthesis of LacI from pLux. As Lac accumulates, it represses pLac which controls the synthesis of LuxI.
