Dephosphorylation
total volume: 50 µl
43 µl plasmid
1 µl SAP (shrink alkaline phosphatase)
5 µl SAP buffer (1:10)
Incubate for 30 minutes at 37°C.
Add one more 1 µl SAP.
Incubate for 20 minutes at 70°C (heat shock).
