An abstraction of this system:

  1. A cell with multi-stable states.
  2. Each stable state has a memory element.
  3. When triggered by outside excitation, the cell goes to another state (decided by memory element).

杨一帆:同上,最好能换一种诱导方式,而不是UV。

刘书摹:这个idea有意思,如果能做成功是一个很有新意的工作。但是设计细节需要仔细斟酌。可能出现的问题是:1,UV照射对细菌的状态有全局影响,细菌的反应可能出现混乱;2,LacI是一个很强的抑制子,光靠分裂稀释可能不足以去除其作用;3,CI不是一个好的转录激活因子,不一定能用;4,两个反向转录而相互抑制的启动子,可以考虑使用phage186。

倪鸣:使用UV照射的一个原因是LexA的转录调控很清楚,会给我们的设计带来很多方便。

Inspiration we get from Drew Endy's story of electric circuit: The naturally evolved bio-systems are not for understanding of human beings. We should design our own bio-systems.

POPS is a key idea from iGEM.

About project idea:

  1. It must be COOL.
  2. There should be progressive milestones. In the last 2 years, no more than 10% of the teams had biologically feasible products when they went to the Jumboree.

用RNA实现基因调控。利用适配子与RNA自催化酶(RNA自催化酶是一段具有斧头形二级结构的RNA,能催化自身降解)组合成调控元件。适配子结合了特定的小分子后,二级结构改变,进而影响RNA自催化酶的二级结构,触发其降解,由此可调控mRNA的水平。利用类似原理还可以控制RNAi过程。
