5:30 Action points from last meeting:

• Alisa: Wiki "Parts_used status" up to date? -(outstanfding from 19/7/07)
• Jan: Lecture Timetables spread sheet complete: attached.
• Phil: Status of #BBa-M30109 & #BBa-V1012 = "will ship this week".
• Jan: Production of sequence for blue light receptor? " -designed

5:45 Status Reports: (5 minutes each including any discussion)

• Alisa: Ligation experiments progress.
  • Problem: 1.2KB genes between biobrick forward & reverse primers not being visualised in colony PCR with 1.5min extn time. (300bp fragments do amplify)
    • - craig to try and repeat.
    • - Alisa: has digestion in progress as alternative diagnostic to prove if problem is ligation or PCR.
    • - Alisa: believes alkaline phosphatase damage is cause of problem.
• Alisa: Red light receptor progress/design/plan- needs.
  • Plan to be completed to be put on wiki this week, estimated at six weeks duration.
  • Phil would like to see lacZ coliphotography parts available for tests due to higher sensitivity of enzyme output.
• Jan: Blue light receptor design/plan - needs.
  • design completed, Plan to be completed and put on wiki this week -Jan
  • Theory to be put on the wiki this week - Jan,-Alisa to review
  • ComA to be ordered by tuesday under discount plan
  • ComP fusion to be ordered ASAP.

• Phil: Gas vesicles progress/design/plan - needs.
  • Plan up to basic biobrick complete,
  • Primers not requested for Friday delivery so cant proceed this week end.
  • Mutagenesis primers will be redesigned by hand for delivery next Friday.
  • Biobrick prefix/sufix primers to be designed this week.
  • Plan for support parts/test harness required.
  • Some items require ordering,round bottom falcon tubes, or manufacture NZY broth, blue/white plate.
  • Lac promotor and antibiotic death plasmids required.
• Pat:
  • Will transform, growup and prepare glycerol stocks of death plasmid in DB3.1 this week.
• Kasia: Documentation progress vs last report/ new problems/ needs.-No report
