Recombination Rate Test
We obtained clones on every plate ! cool. Our competent bacteria were well competent...
Then we took 100 FX85 clones and we put them on AMP-KAN-THYMINE plates in order to determine how many of them lose their kan resistance.
We dillute to 10-7 our liquid culture and then we spread 100µL onto AMP-THYMINE plates.
Transformation of W121 with DapAp-mRFP constructrion
We obtained clones from our last transformation. We took one clone from the plate with L26.1 transformation and 1 from L26.2 transformation.
Then we spread in 5mL of LB those clones with Amp+erythro+DAP.
