AHL assay
Purpose1:
To see how precisely we determine the fluorescence from GFP by removing LB media.
Samples:
placQI + GFP in pTrc99A (pos. con.)
A4 ΔP+ GFP (neg. con.)
Procedure:
AHL assay Standard protocol
Make 200 ul of mixtures of pos. con. and neg. con.
The mixture ratios of pos. con. and neg. con. are
(100:0), (70:30), (50:50), (30:70), (10:90), (3:97), (1:99)
Wash
Result & conclusion:
In proportion to the percentages of GFP expressing cells, fluorescence was increased as shown in Fig. 1.
This indicates that the "Wash
" process well removes the other factors influencing the fluorescence.
