  • One unit is defined as the amount of enzyme required to hydrolyze 1µg of λ DNA to completion in 60 minutes in a total reaction volume of 50 µl, under optimal assay conditions (salt concentration, pH, temperature).
  • Some enzymes which do not, or rarely, cleave λ DNA. DNA are tested using another substrate; for example, enzymes such as Not I or Sfi I are tested with adenovirus DNA, and Fok I is tested using pBR322.
