5:45 Status Reports: (5 minutes each including any discussion)
- Alisa: Ligation experiments progress.
- Problem: 1.2KB genes between biobrick forward & reverse primers not being visualised in colony PCR with 1.5min extn time. (300bp fragments do amplify)
- - craig same ligation worked fine [no BSA, no gel purification, no Alkaline phosphatase used]
- - craig very high 10mg/ml DNA yield from miniprep using alkaline lysis technique.
- - Alisa: doesn't believe it is worth investigating any further except as experiments are done.

- phase 1 osmolarity test of YFP under enz

- Phil sugests using prism for optimal colour discovery before ordering filters, Projector as source.

- Plan up to basic biobrick complete,
- First mutagenesis in progress using complementry primers hand designed, and one overlapping and one computer generated using stratagene program.
- Plan for support parts/test harness required.
- Lac promotor and antibiotic death plasmids required.
- Kasia: Documentation progress vs last report/ new problems/ needs.-No report
