AHL Expression Test
BiobrickのAHLsender&recieverを使ってシグナル伝達してるかテスト.

- 条件
- Strain:XL10
- 液体培地(LB-Amp)
- 固体培地(LB-Amp/1.5% Agar)
- SwarmPlate(Triptone-Amp/0.25% Agar)
- 実験結果
- (福富くんに聞く)
- Sender2・[http://partsregistry.org/Part:BBa_S03623 BBa_S03623] (TetP-LuxI) の方がAHL放出量が多い
- 使ったE.coli(Strain:XL10) 内にLacR有がTetR無い
- よってLacPはある程度抑制されているがTetPは抑制されていない
- Notes
- XL10はAHLをあまりつくらないことを後から知った.田代さん達の実験データよりBW25113とかHB101が良い
8/17(Thu)
AHL Expression Test その2
以下のように条件を変えて8/16と同様にAHL伝達テスト.
- プレートを薄くした
- Senderを濃くした(遠心)
- Senderの濃度をあげてもある程度(どれくらい?)でAHLの濃度は収束する
- Recieverの濃度ふった
- Recieverの濃度が高すぎるとAHL生成量(AHL濃度?)が少なくなる
- 次やること
- Met(AHLのpre-precursur)加えたプレートでAHL生成量増えるかテスト
- Agarの濃度ふる
- Recieverの濃度ふる
